Earth Ponies Playing American Football? Normal show version of the ponies. Not OP fan fiction version.
Still though. If we going the OP route. Do think it could end in disaster? With many humans end up badly hurt during the game... if not outright killed. The ponies end up healing the broken human bodies with magic afterward and bringing back to life the humans they accidentally killed. And Princess Celestia declares any rough contact sport with humans and ponies together is banned. Other than gentle cuddle wrestling.
I feel like this could be a funny story. Imagine human and ponies watching the game turn into a bloodbath as the ponies break the bones and end up literally killing the other team with OPness.
I picture the result being something akin to this:

for a story, you could also go the route, that instead of humans playing against ponies, the humans just watch the ponies play against each other.
If its an anthropomorphic pony world, the humans could be expecting just a sexier version of the regular sport, with giant butts and boobs, only to shit their pants, once they realize, how powerful the ponies are.
